The first flight of Syrian Airlines from Damascus landed at 19:17 local time (16:17 GMT) at Istanbul Airport, where a reception was held for the crew.
SANA quoted Syrian Airlines Director General Sameh Arabi as saying, "Work is underway to expand the flight route to include Turkish cities such as Mersin, Antakya, and Izmir."
Syrian Airlines is scheduled to operate five weekly flights between Damascus and Istanbul.
Turkish Airlines, the national carrier, resumed flights to Damascus in January, six weeks after the Syrian regime was toppled.
Western sanctions were imposed on Syrian Airlines, the national airline, in 2011. Last month, the United States and the European Union lifted economic sanctions on Syria, enabling the government and private sector to reactivate relations with regional and international financial institutions after years of isolation.
