Venus Williams received a wildcard to play doubles with former US Open finalist Leylah Fernandez after losing in the mixed doubles tournament with Reily Opelka.
In singles, Williams lost her first-round match against 11th seed Karolina Muchova, while Fernandez remains in the singles draw.
The seven-time Slam singles champion felt the support of the New York crowd at Arthur Ashe Stadium.
"I don't think I've ever had that much support," Williams was quoted as saying by the WTA.
"When I lost the first two games, they were always by my side every time I won a point. It didn't matter if I was losing and things started going fast, they were always by my side. It felt amazing."
